CF-028 is a small molecule positive allosteric modulator (PAM) of the A3 adenosine receptor (A3AR), developed by Can-Fite BioPharma. Unlike orthosteric agonists, CF-028 enhances the affinity and efficacy of the endogenous ligand (adenosine) specifically at the A3AR subtype. This mechanism is intended to provide a more localized and regulated therapeutic effect, as adenosine levels are typically elevated in pathological tissues such as inflammatory or cancerous sites. CF-028 has been investigated in preclinical models for its potential anti-inflammatory and anti-cancer properties, particularly in melanoma, where it has shown additive effects when combined with other A3AR-targeted therapies.
