CF33 is a novel chimeric oncolytic poxvirus platform developed at City of Hope and licensed by Imugene. It was created by combining genomic sequences from multiple vaccinia virus strains to produce a potent, safe, and highly selective virus with broad-spectrum anti-tumor activity. CF33 selectively infects and replicates within cancer cells, leading to direct cell lysis and the release of tumor-associated antigens, which subsequently stimulates a systemic anti-tumor immune response. The platform is highly modular and has been engineered into several clinical-stage derivatives, including Vaxinia (CF33-hNIS), which expresses the human sodium iodide symporter for imaging and potential radiotherapy, and CheckVacc (CF33-hNIS-anti-PD-L1), which incorporates an immune checkpoint inhibitor. It is currently being evaluated in Phase 1 clinical trials for various advanced solid tumors, both as a monotherapy and in combination with other agents or delivery technologies like RenovoRx's Trans-Arterial Micro-Perfusion (TAMP).
