Overview

CF33-CD19 + blinatumomab is a combination therapy under clinical investigation for advanced or metastatic solid tumors. CF33-CD19 is a novel chimeric oncolytic poxvirus that selectively infects and lyses tumor cells and has been engineered to express a truncated, non-signaling form of human CD19 on the surface of infected tumor cells. This viral tagging enables the use of CD19-directed therapies, including bispecific T-cell engagers such as blinatumomab. Blinatumomab is a bispecific monoclonal antibody that binds CD19 and CD3, activating T cells to recognize and destroy CD19-expressing tumor cells. This combination aims to enable "targetless" solid tumors to be attacked by T cells through synthetic CD19 expression and BiTE-mediated T cell redirection. The combination is in phase I dose escalation/expansion trials with intravenous (IV) and intratumoral (IT) administration, with endpoints including safety, tolerability, and demonstration of anti-tumor activity[1][2][4][5][7][9].
