Overview

CF33-hNIS-anti-PD-L1 is a novel chimeric oncolytic orthopoxvirus engineered to express two transgenes: the human sodium iodide symporter (hNIS) and an anti-programmed death-ligand 1 (PD-L1) single-chain variable fragment antibody. This design enables the virus to selectively infect and lyse tumor cells while simultaneously delivering local immune checkpoint inhibition by blocking PD-L1 within the tumor microenvironment. The hNIS transgene allows for noninvasive imaging and potential radiotherapy applications. The anti-PD-L1 component reverses T-cell inactivation caused by PD-1/PD-L1 signaling, enhancing cytotoxic T lymphocyte-mediated antitumor responses against PD-L1-expressing tumors. Developed primarily for solid tumors, including metastatic triple-negative breast cancer (mTNBC), pancreatic ductal adenocarcinoma (PDAC), and gastric cancer peritoneal metastases, it is currently under investigation in phase I clinical trials as both monotherapy and in combination with other agents[1][2][3][4][5].
