CFI-402411 is an orally available, highly potent small molecule inhibitor of hematopoietic progenitor kinase 1 (HPK1; also known as mitogen-activated protein kinase kinase kinase kinase 1 or MAP4K1), a serine/threonine protein kinase predominantly expressed in hematopoietic cells[1][4][5]. HPK1 acts as a negative regulator of T-cell receptor signaling and immune cell activation. By inhibiting HPK1, CFI-402411 is designed to relieve this inhibition, thereby enhancing T-cell and B-cell function and promoting anti-tumor immune responses[4][5][8]. Preclinical studies have shown that it can disrupt abnormal cytokine expression, alter the tumor immunosuppressive environment (including effects on regulatory T cells), and demonstrate anti-leukemic activity in mouse models[5][6]. The drug is being developed primarily for use in advanced solid malignancies both as monotherapy and in combination with pembrolizumab (an anti-PD-1 antibody)[3][6].
