CFM-4.17 is a small molecule functional mimetic of the Cell Cycle and Apoptosis Regulatory Protein 1 (CARP-1), also known as CCAR1. CARP-1 is a peri-nuclear phosphoprotein that plays a critical role in regulating cell growth, signaling, and apoptosis. CFM-4.17 is designed to mimic the pro-apoptotic functions of CARP-1, which are often downregulated in cancer cells. In preclinical studies, particularly for non-small cell lung cancer (NSCLC), CFM-4.17 has demonstrated the ability to inhibit tumor growth and overcome resistance to EGFR tyrosine kinase inhibitors like osimertinib. Its mechanism involves the modulation of multiple signaling pathways, including the downregulation of STAT3, NF-κB, and PD-L1, and the activation of AMPK. It is frequently studied in nanoformulations (referred to as CFM-F) to enhance its therapeutic profile and delivery in experimental models.
