Overview

CFT-2718 is a **small molecule PROTAC degrader** targeting **BRD4** (bromodomain-containing protein 4), with enhanced catalytic and in vivo activity compared to earlier compounds. BRD4 is a member of the BET (bromodomain and extraterminal) protein family and acts as a core transcriptional and epigenetic regulator involved in recruiting transcriptional complexes to RNA polymerase II. By promoting selective degradation of BRD4, CFT-2718 leads to suppression of transcriptional elongation—particularly affecting cancer cells dependent on super-enhancer-driven transcription. Preclinical studies demonstrate efficacy of CFT-2718 in reducing cell viability and tumor growth in **small-cell lung cancer (SCLC)** and **pancreatic cancer** models. CFT-2718 distinguishes itself from traditional BRD4 inhibitors by its mechanism of catalytic degradation, leading to rapid and potent ablation of BRD4 protein, and has demonstrated greater or comparable efficacy to the CDK9 inhibitor dinaciclib in xenograft models.
