Overview

CG-7321 is an orally bioavailable small molecule inhibitor targeting cyclin-dependent kinases 4 and 6 (CDK4/6), developed by Chengdu Saijing Biomedical Technology. By selectively inhibiting CDK4 and CDK6, the drug prevents the phosphorylation of the retinoblastoma (Rb) protein, thereby blocking cell cycle progression from the G1 to the S phase. This mechanism is particularly effective in treating hormone receptor-positive (HR+) and human epidermal growth factor receptor 2-negative (HER2-) breast cancers, where the CDK4/6-cyclin D-Rb pathway is frequently overactive. CG-7321 is currently undergoing Phase 1 clinical evaluation for the treatment of advanced solid tumors and breast cancer.
