Overview

CG1-CTL refers to an experimental cellular immunotherapy consisting of cytotoxic T lymphocytes (CTLs) that specifically target the CG1 peptide, an HLA-A*0201-restricted nonameric epitope derived from the myeloid protease cathepsin G (CG). Cathepsin G is an azurophil granule protease that is highly expressed and aberrantly processed in acute myeloid leukemia (AML) blasts and leukemia stem cells. In these malignant cells, CG is localized in compartments that facilitate antigen presentation on the cell surface via MHC class I molecules. CG1-CTLs demonstrate selective cytotoxicity against primary AML cells while sparing normal bone marrow, suggesting a favorable safety profile for targeted therapy. Functional CG1-CTLs have been identified in the peripheral blood of AML patients who achieve remission following allogeneic stem cell transplantation, indicating their role in the natural graft-versus-leukemia effect and their potential for further development as a therapeutic agent to prevent or treat relapse.
