Overview

CG5757 is an oncolytic adenovirus developed by Cell Genesys for the treatment of various solid tumors. It is a genetically engineered biologic designed to selectively replicate in and destroy cancer cells while sparing normal tissue. The virus's selectivity is achieved through a dual-promoter system: the E2F-1 promoter, which targets cells with defective retinoblastoma (Rb) pathways, and the human telomerase reverse transcriptase (hTERT) promoter, which targets cells with upregulated telomerase activity. These promoters control the expression of the essential viral genes E1a and E1b. Additionally, CG5757 contains a deletion in the E1B 19k gene to enhance its cytotoxic potential. Preclinical studies have demonstrated efficacy in multiple tumor types, including hepatocellular carcinoma, colorectal cancer, lung cancer, pancreatic cancer, bladder cancer, and cervical cancer.
