Overview

CGC729 is an autologous anti-CD70 chimeric antigen receptor natural killer T (CAR-NKT) cell therapy developed by Cure Genetics for the treatment of relapsed or metastatic advanced renal cell carcinoma and other solid tumors. The therapy utilizes NKT cells genetically engineered via CRISPR-Cas gene editing to express a CAR targeting CD70, a protein frequently expressed in renal cell carcinoma. CGC729 combines adaptive and innate immune mechanisms, enabling multiple tumor-killing actions by targeting CD70, CD1d, and stress ligands. It also modulates the tumor microenvironment (TME) by skewing macrophage differentiation and resisting TGF-β inhibition. Interim Phase I clinical trial data show promising safety and efficacy profiles, with notable response rates in patients with advanced disease[1][2][5][6][7].
