CGEN-54 is a novel, soluble splice variant of the B7-H3 (CD276) immune checkpoint protein, discovered by Compugen using its proprietary computational discovery platform. B7-H3 is a member of the B7/CD28 family of costimulatory molecules that typically provides inhibitory signals to T-cells, thereby facilitating immune evasion by tumors. CGEN-54 was identified as a naturally occurring, secreted form of this protein. Compugen explored CGEN-54 both as a potential therapeutic agent (in the form of an Fc-fusion protein) for autoimmune diseases and as a target for monoclonal antibody development in oncology. Despite its initial promise as a diagnostic marker and therapeutic target, CGEN-54 does not appear in Compugen's current clinical-stage pipeline, suggesting that its development has been discontinued or is no longer a primary focus.
