Drug intelligence / Profile preview
CGSPGWVRC-GG-VLRRDKYGPPT is a lung endothelial-targeted inhibitory peptide designed to block the lactylation of serine and arginine-rich splicing factor 4 (SRSF4) at the lysine 95 (K95) site. Developed by researchers at Fudan University and Zhongshan Hospital, the peptide consists of a lung-homing motif (CGSPGWVRC) linked via a gly-gly (GG) spacer to an inhibitory sequence targeting SRSF4 K95 lactylation. In preclinical models of acute lung injury (ALI) and acute respiratory distress syndrome (ARDS), the peptide prevents SRSF4 K95 lactylation, thereby suppressing the alternative splicing of acid sphingomyelinase (encoded by SMPD1) to the active ASM-1 isoform. This inhibition reduces ceramide production, attenuates vascular barrier dysfunction, and mitigates pulmonary inflammation and vascular permeability.
