Overview

CH-1504 is an orally administered, metabolically inert antifolate small molecule developed for the treatment of rheumatoid arthritis and other immune-mediated diseases. It acts as a **dihydrofolate reductase (DHFR) inhibitor** and is structurally related to methotrexate but designed to avoid the formation of toxic metabolites associated with classical antifolates. In clinical studies, CH-1504 demonstrated comparable efficacy to methotrexate in patients with moderate to severe rheumatoid arthritis while showing improved safety and tolerability profiles. The drug was originally developed by Chelsea Therapeutics under license from the University of South Alabama; Chelsea Therapeutics was later acquired by Lundbeck. Development for indications including rheumatoid arthritis, cancer, inflammatory bowel diseases, and psoriasis has been discontinued[1][3][5][8].
