Overview

ch128.1 is a mouse/human chimeric IgG3 monoclonal antibody that targets the human transferrin receptor 1 (TfR1), also known as CD71. Developed by researchers at UCLA, the antibody is designed to treat hematopoietic malignancies, particularly multiple myeloma. Its mechanism of action involves direct cytotoxicity through the induction of TfR1 degradation and subsequent lethal iron deprivation in malignant cells. Additionally, ch128.1 elicits Fc-mediated effector functions, including antibody-dependent cell-mediated cytotoxicity (ADCC) and antibody-dependent cell-mediated phagocytosis (ADCP), primarily mediated by macrophages in vivo. While it can bind C1q and induce complement-mediated cytotoxicity (CDC) in vitro, studies suggest CDC may not be the primary driver of its anti-tumor activity in xenograft models.
