ch128.1 Fc mutant is a modified version of the mouse/human chimeric IgG3 antibody ch128.1, which targets the human transferrin receptor 1 (TfR1), also known as CD71. This specific mutant is engineered with mutations in the Fc region to impair its binding to Fc gamma receptors (FcγRs) and the complement component C1q, thereby abrogating its ability to trigger antibody-dependent cellular cytotoxicity (ADCC), antibody-dependent cellular phagocytosis (ADCP), and complement-dependent cytotoxicity (CDC). In preclinical research, particularly in xenograft models of multiple myeloma, this mutant serves as a critical tool to demonstrate that the in vivo efficacy of the parent ch128.1 antibody is dependent on host effector functions rather than solely on direct cytotoxicity via iron deprivation or receptor degradation.
