**ch225-IL2** is an experimental EGFR-targeted immunocytokine consisting of a mouse-human chimeric anti-epidermal growth factor receptor antibody derived from mAb 225, with human interleukin-2 genetically fused to the antibody heavy-chain C-terminus. The antibody component targets EGFR-expressing tumor cells and can block ligand-driven EGFR signaling, while the IL-2 payload activates IL-2 receptor-expressing immune effector cells, including activated T cells and natural killer cells. In preclinical human melanoma xenograft models reconstituted with human lymphokine-activated killer cells, ch225-IL2 eradicated established hepatic and pulmonary micrometastases and improved survival relative to separately administered antibody plus recombinant IL-2.
