Overview

CH3127 is an oral prodrug of semaglutide, a glucagon-like peptide-1 (GLP-1) receptor agonist, developed for once-weekly administration. Developed by Chuanghui Biotechnology, the molecule is engineered to act as an in vivo reservoir that releases semaglutide at a controlled rate, resulting in a pharmacokinetic profile characterized by a reduced peak concentration (Cmax) and an increased trough concentration (Ctrough). This "flat" drug concentration-time curve is intended to enhance therapeutic efficacy and improve tolerability by minimizing the gastrointestinal side effects typically associated with high Cmax levels of GLP-1 receptor agonists. CH3127 has demonstrated high oral bioavailability in non-human primates and has received FDA Investigational New Drug (IND) clearance for clinical evaluation in metabolic disorders such as type 2 diabetes and obesity.
