ch3D6kLALA is a chimeric research antibody construct targeting amyloid beta (Aβ), derived from the 3D6 antibody. It is engineered with asymmetric Leucine-to-Alanine (LALA) mutations in the human IgG Fc region to reduce effector function and mitigate amyloid-related imaging abnormalities (ARIA), such as microhemorrhages and edema, which are common side effects of anti-Aβ immunotherapies. Developed by Denali Therapeutics in collaboration with Biogen, this construct serves as a foundational molecule for testing the safety and efficacy of Fc-engineered antibodies and is often compared against Antibody Transport Vehicle (ATV) modified variants that target the transferrin receptor (TfR) for improved blood-brain barrier penetration in Alzheimer's disease models.
