ChAd155-hIi-HBV + MVA-HBV is a therapeutic vaccine combination developed by GSK for the treatment of chronic hepatitis B (CHB). The regimen utilizes a heterologous prime-boost strategy designed to induce robust B-cell and T-cell responses against the hepatitis B virus. The prime component, ChAd155-hIi-HBV, is a replication-defective chimpanzee adenovirus (serotype 155) vector encoding the HBV core (HBcAg) and surface (S-HBsAg) antigens, with the core antigen fused to the human MHC class II-associated invariant chain (hIi) to enhance immunogenicity. The boost component, MVA-HBV, is a modified vaccinia Ankara vector encoding the same antigens. This combination is frequently evaluated as part of the GSK3528869A clinical program, which also includes an adjuvanted protein vaccine (HBc-HBs/AS01B). The primary goal of the therapy is to achieve functional cure in patients with CHB by reducing HBsAg levels and restoring immune control over the virus.
