Overview

ChAd155-hIi-HBV vaccine is an investigational recombinant chimpanzee adenovirus type 155-based viral vector vaccine developed by GSK for the treatment of chronic hepatitis B (CHB). The vaccine construct utilizes a replication-defective chimpanzee adenovirus serotype 155 (ChAd155) backbone encoding two hepatitis B virus (HBV) antigens: the truncated core nucleocapsid protein antigen (HBc) and the full-length small surface antigen (HBs). To enhance antigen presentation and stimulate a robust T-cell and antibody immune response, the human MHC class II-associated invariant chain (hIi, CD74, p35 isoform) is fused to the HBc protein. ChAd155-hIi-HBV has been evaluated in clinical trials as the 'prime' component of a heterologous prime-boost immunotherapy regimen (followed by MVA-HBV and adjuvanted protein vaccines) and in sequential combination with the antisense oligonucleotide bepirovirsen.
