ChAd63-MVA AMA1 is an experimental viral-vectored malaria vaccine developed by the University of Oxford. It targets the blood-stage of *Plasmodium falciparum* infection by expressing the apical membrane antigen 1 (AMA1), a merozoite surface protein essential for red blood cell invasion. The vaccine utilizes a heterologous prime-boost strategy: a chimpanzee adenovirus 63 (ChAd63) vector is used for priming, followed by a modified vaccinia virus Ankara (MVA) vector for boosting, typically administered eight weeks apart. This regimen is designed to elicit robust cellular (CD4+ and CD8+ T cells) and humoral (antibody) immune responses. While Phase I/IIa clinical trials, including controlled human malaria infection (CHMI) studies, have demonstrated the vaccine's safety and immunogenicity, it has shown limited efficacy against blood-stage parasitemia as a standalone candidate.
