Overview

ChAd63-PfCAT is a recombinant, non-replicating simian adenovirus serotype 63 (ChAd63) vectored vaccine candidate developed by the Jenner Institute at the University of Oxford for the prevention of malaria. The vaccine encodes a unique fusion protein, PfCAT, which consists of three highly conserved *Plasmodium falciparum* antigens: the circumsporozoite protein (CSP), apical membrane antigen 1 (AMA1), and thrombospondin-related anonymous protein (TRAP). This multi-antigen approach is designed to target the malaria parasite at both the pre-erythrocytic stage (via CSP and TRAP) and the blood stage (via AMA1). By utilizing a viral vector to deliver the genetic sequence for these antigens, the vaccine induces robust cellular (CD8+ and CD4+ T-cell) and humoral (antibody) immune responses. In clinical development, ChAd63-PfCAT is frequently evaluated in heterologous prime-boost regimens alongside a Modified Vaccinia Virus Ankara (MVA) vector encoding the same antigens (MVA-PfCAT) to enhance protective efficacy.
