Overview

ChAd63 PvDBP is a recombinant viral-vectored vaccine candidate designed to prevent malaria caused by Plasmodium vivax. It uses a replication-defective chimpanzee adenovirus serotype 63 (ChAd63) as the prime vector to deliver the gene encoding region II of the P. vivax Duffy-binding protein (PvDBPII), a critical domain involved in parasite invasion of human red blood cells via interaction with the Duffy Antigen Receptor for Chemokines (DARC). The vaccine is typically administered in a prime-boost regimen with MVA PvDBP as the booster. Clinical trials have shown that this approach induces both antibody and T cell responses against PvDBPII, generating strain-transcending antibodies capable of inhibiting parasite binding to DARC and reducing parasitemia after controlled human malaria infection[1][2][3][4][6][7].
