ChAdOx1-triFilo is a trivalent, recombinant viral vector vaccine candidate developed by the University of Oxford to provide broad protection against major filoviruses. It utilizes a replication-deficient chimpanzee-derived adenoviral vector (ChAdOx1) engineered to encode and express the glycoproteins (GP) of three distinct viruses: Zaire ebolavirus (EBOV), Sudan ebolavirus (SUDV), and Marburg virus (MARV). Unlike many multivalent vaccine strategies that involve mixing separate monovalent vectors, ChAdOx1-triFilo is designed as a single-vector system expressing all three antigens, which may streamline manufacturing and reduce costs. The vaccine is intended to induce both humoral and cellular immune responses against the viral glycoproteins, which are the primary surface proteins responsible for viral entry into host cells. Preclinical studies in guinea pig models have demonstrated protective efficacy against lethal challenge, and the platform is being advanced for outbreak preparedness in endemic regions.
