ChAdOx1 Zika is a recombinant viral vector vaccine candidate developed to prevent infection by the Zika virus. It uses the non-replicating chimpanzee adenovirus vector platform (ChAdOx1), engineered to express structural proteins from the Zika virus—most notably, either the envelope (E) protein or a combination of pre-membrane and envelope (prM-E) proteins. The vaccine is designed to induce both humoral and cellular immune responses against ZIKV antigens. In preclinical studies, single-dose immunization with constructs such as ChAdOx1 prMEΔTM has demonstrated protection in mouse models by clearing viremia and reducing viral load in tissues; it also elicited strong antibody and T-cell responses. The mechanism of action involves delivery of genetic material encoding key viral antigens into host cells via the adenoviral vector, leading to antigen expression and subsequent immune activation targeting those antigens[3][5][6]. Developed at the Jenner Institute at University of Oxford, this vaccine represents an effort to provide broad protection against multiple lineages of ZIKV[4][5].
