ChAdOx1.HTI + MVA.HTI is a therapeutic HIV vaccine regimen consisting of two viral vector-based biologics designed to induce a potent and broad T-cell immune response against HIV-1. Developed by AELIX Therapeutics (acquired by Gilead Sciences), the regimen utilizes the HIVACAT T-cell immunogen (HTI), which is engineered to target the most vulnerable and highly conserved regions of the HIV-1 proteome to prevent viral escape. The regimen follows a heterologous prime-boost strategy: ChAdOx1.HTI, which uses a chimpanzee adenovirus Oxford 1 vector, is administered as the prime dose, followed by MVA.HTI, which uses a modified vaccinia Ankara vector, as the boost. The goal of this therapy is to achieve a 'functional cure,' enabling HIV-infected individuals to maintain viral suppression without the need for lifelong antiretroviral therapy (ART). Clinical trials, such as AELIX-002 and AELIX-003, have evaluated this regimen both alone and in combination with other agents like the TLR7 agonist vesatolimod.
