ChAdOx2 RabG is a novel simian adenovirus-vectored vaccine designed for cost-effective pre-exposure prophylaxis against rabies. It utilizes a modified chimpanzee adenovirus serotype 68 (AdC68) backbone, enhanced by incorporating the human adenovirus serotype 5 (AdHu5) E4 orf6/7 region in place of the AdC68 equivalents. This modification facilitates manufacturing in cell lines providing AdHu5 E1 proteins in trans. Developed at the Jenner Institute, the vaccine is unable to replicate in mammalian cells while still producing an effective immune response. It's designed as a single-dose option, particularly targeting low and middle-income countries where rabies remains a significant public health concern. The vaccine has shown high immunogenicity in animal studies, inducing antibody responses comparable to other adenovirus-vectored rabies vaccines and achieving high titers of rabies virus neutralizing antibody (VNA) after a single dose. It also demonstrates thermostability using a low-cost method suitable for ambient-temperature distribution in tropical climates and has dose-sparing potential when formulated with a simple chemical adjuvant.
