Overview

ChAdV63.HIVconsv is an investigational therapeutic HIV vaccine designed to elicit strong T-cell responses against highly conserved regions of the HIV-1 proteome. The vaccine uses a non-replicating chimpanzee adenovirus serotype 63 (ChAdV63) vector to deliver a chimeric immunogen, termed HIVconsv, which is constructed from 14 of the most conserved regions across multiple clades (A, B, C, D) of HIV-1. This approach aims to focus immune responses on subdominant but highly conserved viral epitopes that are less likely to mutate and escape immune detection. The primary mechanism is immunostimulation—specifically inducing robust CD8+ cytotoxic T lymphocyte responses capable of inhibiting replication of diverse HIV-1 variants in vitro and in vivo. ChAdV63.HIVconsv has been evaluated as part of prime/boost regimens with MVA.HIVconsv in both prophylactic and therapeutic settings for people living with or at risk for HIV infection[4][6][8]. It has shown safety and immunogenicity in early-phase clinical trials.
