ChAdV68-S is an investigational viral vector vaccine developed by Gritstone bio for the prevention of COVID-19. It utilizes a non-replicating chimpanzee adenovirus vector, ChAdV68, to deliver the genetic instructions for the SARS-CoV-2 spike protein. The use of a chimpanzee adenovirus is intended to circumvent the issue of pre-existing immunity to common human adenoviruses, which can limit vaccine effectiveness. ChAdV68-S is being evaluated in clinical trials, including those sponsored by the National Institute of Allergy and Infectious Diseases (NIAID), as part of heterologous prime-boost vaccination strategies. In these regimens, it is often paired with self-amplifying mRNA (SAM) vaccines to elicit a robust and durable immune response, characterized by both high titers of neutralizing antibodies and potent T-cell activation.
