CHC shRNA is a short hairpin RNA (shRNA) therapeutic candidate designed to silence the expression of clathrin heavy chain (CHC). Research conducted at Academia Sinica has identified CHC as a critical regulator of tumor growth and angiogenesis, beyond its traditional role in endocytosis. The mechanism involves CHC binding to and stabilizing the transcription factor subunit HIF-1α, facilitating its nuclear translocation and subsequent upregulation of VEGF expression. By knocking down CHC, this shRNA disrupts the HIF-1α/VEGF signaling axis, leading to inhibited tumor cell proliferation, colony formation, and vessel formation. Preclinical studies in NOD/SCID mice bearing pancreatic cancer xenografts have demonstrated that CHC shRNA can effectively inhibit tumor progression and angiogenesis.
