Overview

CHC2014 is an investigational, orally available, highly selective pan-tropomyosin receptor kinase (pan-TRK) inhibitor being developed as a targeted anticancer therapy for tumors harboring NTRK gene fusions and resistance-associated TRK mutations. It selectively inhibits TRK proteins and has demonstrated strong and selective antitumor activity in tumor cell lines and animal models, with superior in vivo antitumor efficacy compared with existing treatment options and robust activity against solvent-front and gatekeeper TRK mutations, suggesting potential to overcome acquired resistance to first-generation TRK inhibitors.[2][6][7] Initially co-developed by Handok, CMG Pharmaceutical, and NOV, global (ex-Korea) rights were later licensed to AUM Biosciences, which is advancing CHC2014 in a tumor-agnostic development strategy following a successful first-in-human Phase 1 trial in Korea.[2][6][7][13]
