Overview

Chemotherapy + p53 refers to a therapeutic combination strategy in oncology where standard chemotherapeutic agents are administered together with interventions that target or restore the function of the tumor suppressor protein p53. This approach is based on the observation that many cancers harbor mutations or loss of function in the TP53 gene, which encodes p53, a key regulator of cell cycle arrest and apoptosis in response to DNA damage. The rationale for combining chemotherapy (which induces DNA damage) with therapies that restore or enhance p53 activity is to increase cancer cell death, particularly in tumors with defective endogenous p53 pathways. Approaches include using recombinant human p53 adenovirus (rAd-p53) gene therapy alongside chemotherapeutic drugs, aiming for synergistic anti-tumor effects by both damaging DNA and restoring apoptotic signaling[5]. This combination has been explored preclinically and clinically across various solid tumors.
