Overview

CHF-1024 is a cyclized phenethylamine analogue of dopamine and acts as a dopamine D2 receptor agonist and α2-adrenergic receptor agonist.[1][3] It belongs to the 2-aminotetralin chemical family and is structurally related to catecholamines. CHF-1024 is the pharmacologically active form of nolomirole (CHF-1035), which is a prodrug; nolomirole and CHF-1024 were investigated for potential use in cardiovascular diseases such as heart failure, but they were never marketed.[1] Animal studies indicate CHF-1024 can attenuate neuroendocrine activation, blunt norepinephrine excretion, and reduce cardiac fibrosis in pressure-overload hypertrophy, demonstrating effects on cardiac adrenergic drive and potential cardioprotective properties.[3][5]
