Overview

CHF6297 is a novel, potent, and selective small molecule inhibitor of p38α mitogen-activated protein kinase (MAPK14), designed for inhalation as a dry powder formulation. It exhibits sub-nanomolar potency against p38α (IC50 = 0.14 nM) with over 1,000-fold selectivity versus other p38 isoforms. In preclinical studies, CHF6297 demonstrated strong anti-inflammatory effects by inhibiting neutrophil influx and cytokine release in models of lung inflammation where corticosteroids are less effective. The drug has shown efficacy in experimental models relevant to acute exacerbations of chronic obstructive pulmonary disease (COPD), asthma, acute lung injury (ALI), and viral-induced hyperinflammation[1][2][3][4][5]. It is being developed primarily for the treatment of COPD and has also been investigated preclinically for asthma.
