Overview

CHF6467 is a recombinant mutated form of human nerve growth factor (NGF), specifically the R100E variant, developed to retain neuroprotective and neurotrophic properties while significantly reducing pain-inducing activity compared to native NGF. It is being investigated primarily as a topical or intranasal therapy for conditions involving neurodegeneration or impaired wound healing. CHF6467 has shown potential in preclinical and clinical studies for promoting neuronal survival and function without the adverse effect of pain typically associated with NGF therapies. The drug is under development by Chiesi Farmaceutici for indications including diabetic foot ulcers and optic pathway glioma, with additional research in hypoxic-ischemic encephalopathy (HIE) suggesting synergistic effects when combined with therapeutic hypothermia.
