CHI-29B chimera is a **synthetic RNA chimera** composed of a MUC1 aptamer conjugated to microRNA-29b (miR-29b)[1][3]. It is designed to specifically target MUC1-expressing ovarian cancer cells, delivering miR-29b and inducing anti-tumor and anti-chemoresistant effects by epigenetically modulating gene expression. Mechanistically, CHI-29B chimera inhibits tumor growth by downregulating PTEN methylation (resulting in increased PTEN expression), and suppresses the expression of key oncogenic mediators such as MAPK 4, MAPK 10, and IGF1[1][3]. In paclitaxel-resistant ovarian tumor models, the chimera enhances apoptosis through activation of PTEN signaling and inhibition of cancer stem cell activation[1], which has been shown to overcome chemoresistance. The RNA aptamer targeting MUC1 ensures delivery specificity to MUC1+ cells.
