chidamide + rituximab + gemcitabine + dexamethasone + cisplatin

Chidamide + R-GDP is a combination chemo-immunotherapy regimen consisting of the oral histone deacetylase (HDAC) inhibitor chidamide and the R-GDP backbone, which includes rituximab, gemcitabine, dexamethasone, and cisplatin. This combination is being evaluated by Fudan University Shanghai Cancer Center for the treatment of patients with relapsed or refractory diffuse large B-cell lymphoma (DLBCL) who are not candidates for autologous stem cell transplantation. Chidamide, a selective HDAC inhibitor, works by modulating epigenetic pathways to overcome chemotherapy resistance, while rituximab targets CD20-positive B-cells and the chemotherapy components (gemcitabine and cisplatin) induce DNA damage. Clinical data from a Phase II trial (NCT03373019) indicated that the addition of chidamide to R-GDP (C-R-GDP) significantly improves response rates in this difficult-to-treat population.
