Chidamide + dalpiciclib is an investigational oral combination therapy for hormone receptor-positive, HER2-negative (HR+/HER2-) advanced or metastatic breast cancer. Chidamide (also known as tucidinostat) is a selective histone deacetylase (HDAC) inhibitor that modulates gene expression by altering chromatin structure, leading to cell cycle arrest and apoptosis in cancer cells. Dalpiciclib is a cyclin-dependent kinase 4/6 (CDK4/6) inhibitor that blocks cell cycle progression from G1 to S phase by inhibiting CDK4 and CDK6 activity, thereby suppressing tumor cell proliferation. This combination has been studied in patients who have progressed after prior CDK4/6 inhibitor therapy and chemotherapy for advanced breast cancer. Early-phase clinical trials indicate the regimen may offer an alternative treatment strategy with manageable toxicity profiles; the most common grade 3-4 adverse events are neutropenia and thrombocytopenia[1][2][4].
