Chi-GVM is a combination chemotherapy regimen investigated for the treatment of relapsed or refractory peripheral T-cell lymphoma (PTCL). The regimen consists of chidamide (tucidinostat), gemcitabine, vinorelbine, and mitoxantrone hydrochloride liposome. Chidamide is an orally active, subtype-selective histone deacetylase (HDAC) inhibitor that acts as an epigenetic modulator. Gemcitabine is a nucleoside analog that inhibits DNA synthesis, vinorelbine is a vinca alkaloid that disrupts microtubule assembly, and mitoxantrone hydrochloride liposome is an anthracenedione antineoplastic agent that intercalates into DNA and inhibits topoisomerase II. This multi-agent approach is designed to overcome the poor prognosis and limited durability of standard anthracycline-based therapies in patients with mature T-cell malignancies.
