Overview

Chidamide + ivonescimab is an investigational combination therapy consisting of two agents with distinct mechanisms of action. Chidamide (also known as tucidinostat or HBI-8000) is a selective histone deacetylase inhibitor (HDACi) that targets HDAC1, HDAC2, HDAC3, and HDAC10. It modulates epigenetic regulation to induce cell cycle arrest and apoptosis in tumor cells and can enhance immune recognition by upregulating immune-related surface markers. Ivonescimab (AK112/SMT112) is a first-in-class bispecific monoclonal antibody targeting both programmed death-1 (PD-1) and vascular endothelial growth factor A (VEGF-A). By simultaneously blocking PD-1-mediated immunosuppression and VEGF-driven angiogenesis in the tumor microenvironment, it aims to potentiate antitumor immunity while inhibiting tumor vascularization. The combination is under clinical investigation for advanced bone sarcoma and soft tissue sarcoma as a second-line treatment[7][9].
