Overview

This is a combination therapy consisting of three agents: - **Chidamide** (also known as tucidinostat), an oral histone deacetylase (HDAC) inhibitor that induces apoptosis and cell cycle arrest in tumor cells by inhibiting HDAC activity, leading to changes in gene expression. It is approved for relapsed or refractory peripheral T-cell lymphoma and has shown potential in combination with other agents for solid tumors[1][5]. - **Toripalimab**, a humanized monoclonal antibody targeting programmed cell death protein 1 (PD-1), functioning as an immune checkpoint inhibitor to enhance anti-tumor immune responses. It is used primarily in oncology, including melanoma and non-small cell lung cancer[2][3]. - **Paclitaxel**, a microtubule-stabilizing chemotherapeutic agent of the taxane class that inhibits mitosis by preventing microtubule disassembly, leading to apoptosis of rapidly dividing cells. It is widely used for various cancers including breast, ovarian, and lung cancer[7][10]. The combination aims to leverage epigenetic modulation (chidamide), immune checkpoint blockade (toripalimab), and cytotoxic chemotherapy (paclitaxel) for synergistic anti-tumor effects. Clinical trials are ongoing evaluating this regimen as neoadjuvant therapy in low hormone receptor expression, HER2-negative early breast cancer[2][4], among other indications.
