Chikusetsusaponin IVa is a triterpenoid saponin and a major active component found in several medicinal plants, including Panax japonicus, Aralia taibaiensis, and Aralia elata[1][3]. It exhibits diverse pharmacological activities such as anticancer, anti-inflammatory, antioxidant, antithrombotic, and anti-aging effects[1][7][3]. Mechanistically, it has been shown to inhibit cancer cell proliferation and induce apoptosis via multiple pathways, including mitochondrial control (caspase-dependent and -independent), modulation of Bcl-2/Bax expression, and inhibition of the IL-6/STAT3 and NF-κB pathways[1]. It also activates AMP-activated protein kinase (AMPK), improving autophagy and mitophagy, which contributes to its anti-aging effects[3]. Additionally, it inhibits inflammatory cytokine production and oxidative stress by targeting NF-κB and MAPK signaling, and exerts metabolic benefits by improving insulin sensitivity and glucose homeostasis[1][7]. It has shown potential in preclinical models for cancer, diabetes, and aging-related diseases, but clinical development status remains unclear[1][3][7].
