Overview

This drug consists of T cells engineered to express a chimeric antigen receptor (CAR), specifically targeting CD19, along with membrane-bound forms of interleukin-2 (IL-2) and interleukin-12 (IL-12). Developed by **Portal** (Portal Bio), these CAR-T cells are created using a proprietary mechanoporation technology that utilizes a silicon membrane to deliver circular RNA (circRNA) encoding the CAR and cytokines into primary T cells. The membrane-bound cytokines are intended to provide localized stimulatory signals to the T cells, enhancing their persistence, expansion, and anti-tumor activity within the tumor microenvironment while minimizing systemic toxicity associated with soluble cytokines. This approach aims to improve the efficacy of cell therapies in treating malignancies, particularly B-cell cancers, by addressing the limitations of cytokine support and cell engineering efficiency.
