Chimeric PD-1-DAP10 is a preclinical chimeric antigen receptor (CAR) construct designed to repurpose the PD-1/PD-L1 immunosuppressive axis into an activating signal for adoptive cell therapy. The construct consists of the extracellular domain of the Programmed Death-1 (PD-1) receptor fused to the intracellular co-stimulatory signaling domain of DAP10. By binding to PD-1 ligands (PD-L1 and PD-L2) overexpressed on tumor cells, the chimeric receptor triggers T-cell or NK-cell activation and cytokine secretion (notably IFNγ and TNFα) without the inhibitory signaling typically associated with native PD-1. Research led by Amorette Barber and others has demonstrated its efficacy in murine models across various solid tumors and lymphomas, showing enhanced anti-tumor activity and a favorable pro-inflammatory cytokine profile compared to other co-stimulatory domains.
