Overview

Chk1i-a is a novel, blood-brain barrier (BBB)-permeable small molecule inhibitor of checkpoint kinase 1 (Chk1) developed by researchers at the University of Strathclyde. It is being investigated for the treatment of glioblastoma (GBM), particularly in combination with temozolomide (TMZ) or Wee1 inhibitors (such as Wee1i-a). In preclinical studies using patient-derived glioblastoma stem-like cells, Chk1i-a demonstrated potent single-agent activity and strong synergistic effects when combined with TMZ, leading to enhanced cell death and reduction in clonogenicity.
