chKTi-SPP-DM1 is a non-targeting antibody-drug conjugate (ADC) developed by ImmunoGen for use as a negative control in preclinical research. It consists of a chimeric human IgG1 monoclonal antibody directed against soybean trypsin inhibitor (KTi), an antigen not found in human or mouse tissues, conjugated to the cytotoxic maytansinoid DM1 via the cleavable disulfide linker N-succinimidyl-3-(2-pyridyldithio)propionate (SPP). In experimental models, such as those for small cell lung cancer (SCLC), chKTi-SPP-DM1 serves to establish the baseline for antigen-independent toxicity and to confirm that the efficacy of targeted ADCs (like lorvotuzumab mertansine) is due to specific binding to the intended tumor antigen rather than non-specific payload delivery.
