Chlorazolamide magnesium(II) complex is a metal-based sulfonamide derivative designed as a potent carbonic anhydrase inhibitor. It is derived from chlorazolamide (5-(2-chlorophenyl)-1,3,4-thiadiazole-2-sulfonamide). While the parent sulfonamide exhibits poor topical penetration and weak intraocular pressure (IOP) lowering effects when administered as a suspension, the magnesium(II) complex significantly enhances these pharmacological properties. It acts by inhibiting carbonic anhydrase isozymes (CA I, II, and IV), which play a critical role in ocular fluid secretion. In experimental animal models, the complex has demonstrated the ability to effectively lower IOP, making it a candidate for the treatment of glaucoma and ocular hypertension.
