Chlorin e6 + methylene blue is a **combination of two photosensitizer drugs** used in experimental and investigational **photodynamic therapy (PDT)** research. **Chlorin e6** is a type II photosensitizer that primarily generates singlet oxygen upon irradiation, leading to oxidative destruction of targeted cells. **Methylene blue** is a type I photosensitizer and redox-active dye that generates reactive oxygen species (ROS) via electron transfer reactions; it can also directly participate in mitochondrial redox cycling. When combined, they provide simultaneously type I and type II photodynamic effects, which can increase phototoxicity in cancer cells compared to either agent alone. Co-administration of chlorin e6 with methylene blue also prevents rapid reduction of methylene blue to its inactive leucoform under hypoxic conditions and enhances the intracellular fluorescence and retention of both agents.
