Overview

Chloroquine + dehydroepiandrosterone sulphate (DHEAS) is an experimental combination therapy developed by the Université Victor Segalen Bordeaux 2 for the treatment of uncomplicated drug-resistant *Plasmodium falciparum* malaria. Chloroquine is a 4-aminoquinoline antimalarial that functions by entering the parasite's acidic food vacuole and inhibiting the biocrystallization of toxic heme into non-toxic hemozoin, leading to parasite death. Dehydroepiandrosterone sulphate, a naturally occurring steroid hormone, was investigated as an adjunct to chloroquine based on evidence that it may inhibit parasite growth—possibly through the inhibition of glucose-6-phosphate dehydrogenase (G6PD)—and potentially reverse chloroquine resistance. This combination was evaluated in a Phase 3 clinical trial to determine if DHEAS could restore the efficacy of chloroquine in regions where resistance had rendered the monotherapy ineffective.
